Harvest with Massive Muramasa for maximum reach (6 blocks in front, 4 behind, 3 below, 10 above). Muramasa is accessible relatively early in the game from Dungeon Chests. Reforging it to the Massive (+18% size) prefix will set you back anywhere between 3 and 100+ gold. It all depends on luck.

Farm layouts: I switched to horizontal (first screenshot) after getting the Massive Muramasa. Harvesting 4 layers per pass is great. Before the Muramasa, I used the vertical layout shown in the second screenshot.

This farm works with regular and glowing mushrooms.

Edit: The Blade of Grass has an even bigger reach. Though it is considerably slower.
